چکیده
Thermococcus gammatolerans EJ3 is an extremophile archaeon which was revealed as one of the most radioresistant organisms known on Earth, withstanding up to 30 kGy gamma-ray radiations. While its theoretical proteome rather small, T. may enhance toolbox by post-translational modification proteins. Here, we explored extent N?-acetylation lysines. For this, immunopurified with two acetylated-lysine antibodies acetylated peptides resulting from a proteolysis soluble proteins trypsin. The comparison proteomes archaea highlights some common acetylation patterns but only 4 out 26 orthologous found be in both species, are same lysine site. We evidenced that histone B at least different sites (K27 and K36), peptide C-terminus histones A also acetylated. verified trait among Thermococcales after recording data kodakaraensis identifying three sites. This discovery reinforces strong evolutionary link between Archaea Eukaryotes should incentive for further investigation role Archaea. An Intelligent System for Identifying Acetylated Lysine on Histones and Nonhistone Proteins
Lysine acetylation is an important and ubiquitous posttranslational modification conserved in prokaryotes and eukaryotes. This process, which is dynamically and temporally regulated by histone acetyltransferases and deacetylases, is crucial for numerous essential biological processes such as transcriptional regulation, cellular signaling, and stress response. Since the experimental identificati...
متن کاملGenome-Wide Transcriptional Response of the Archaeon Thermococcus gammatolerans to Cadmium
Thermococcus gammatolerans, the most radioresistant archaeon known to date, is an anaerobic and hyperthermophilic sulfur-reducing organism living in deep-sea hydrothermal vents. Knowledge of mechanisms underlying archaeal metal tolerance in such metal-rich ecosystem is still poorly documented. We showed that T. gammatolerans exhibits high resistance to cadmium (Cd), cobalt (Co) and zinc (Zn), a...
متن کاملADP-ribosylation of core histones and their acetylated subspecies.
ADP-ribosylation of core histones was investigated in isolated nuclei of Physarum polycephalum. Core histone species differed in the mode of modification. Whereas ADP-ribosylation of H2A and H2B is sensitive to inhibition by 3-methoxybenzamide, as with most other nuclear acceptor proteins, the modification of H3 and H4 is not inhibited. Cleavage experiments with hydroxylamine indicate a carboxy...
متن کاملDynamically acetylated histones of chicken erythrocytes are selectively methylated.
The relationship between histone acetylation and methylation in chicken immature erythrocytes was investigated. Previous studies have shown that transcriptionally active/competent gene-enriched chromatin fragments are enriched in newly methylated histones H3 and H4. Moreover, newly methylated histone H4 is hyperacetylated. Here, we show that dynamically acetylated histone H4 is selectively enga...
متن کاملAcetylated and non-acetylated flavonol triglycosides from Galega officinalis.
Three flavonol triglycosides kaempferol 3-[2Gal-(4-acetylrhamnosyl)robinobioside], kaempferol 3-(2Gal-rhamnosylrobinobioside) and quercetin 3-(2G-rhamnosylrutinoside) have been isolated from a methanolic extract of Galega officinalis aerial parts. They are reported for the first time in the genus Galega; moreover, the acetylated triglycoside is a new natural product.
